SDF-1α from adipose derived-stem cell increases angiogenesis in acute myocardial infarction

Abstract Body: Acute myocardial infarction (AMI) occurs when blood flow of coronary arteries is blocked, causing damage to the myocardial. In this study, we found the enhanced secretion SDF-1α from mASCs compared to mECs, and investigated the cellular change of rat ECs according to that difference of SDF-1α expression in vitro. Mouse cytokine antibody array was performed to identify which growth factors/cytokines were secreted from mASCs and mECs. Interestingly, we confirmed that SDF-1α was expressed higher in mASCs CM than mECs CM. Understanding cellular changes in the hypoxic environment is important for cell transplantation on ischemic models. We confirmed that without CoCl2 treatment, the protein expression of p-Akt and p-mTOR were significantly increased in rECs treated with mASCs CM, compared to those of not treated with mASCs CM. We investigated one step further by determining whether mASCs transplantation could improve cardiac function and regeneration on AMI rat model. The EFs of the two cell transplantation groups were both significantly improved compared to the Sham group on day 28 of post-transplantation. Interestingly, comparing the two cell transplantation groups, there was a higher EF improvement in the mASCs group than the mECs group. We visualized the mobilization of CXCR4 induced by SDF-1α secreted from transplanted cells, and it showed significantly higher expression of CXCR4 in mASCs group compared to Sham and mECs groups. Our results suggest that mASCs were transplanted into AMI-induced rats to demonstrate the improvement of cardiac function and angiogenesis by paracrine effect of SDF-1α.
